L'Amantadine peut déclencher ou aggraver les symptômes psychotiques. Rarement, les tentatives de suicide et idées suicidaires ont également été signalés, dont certains sont survenues chez des patients n'ayant aucun antécédent de maladie psychiatrique qui prenaient des cours de courte durée de l'amantadine pour la grippe. La thérapie avec l'amantadine doit être administré avec prudence chez les patients présentant des antécédents de troubles psychiatriques, l'instabilité émotionnelle, ou d'abus de substance, et surtout s'ils sont des personnes âgées. Il peut être prudent de s'abstenir de la distribution de grandes quantités de ce médicament chez ces patients, pour éviter de surdosage intentionnel, qui est potentiellement mortelle.
