L'utilisation des oestrogènes est contre-indiqué dans les patients avec des cas non diagnostiqués, de saignements vaginaux anormaux. Prolongée (> 1 an), sans opposition, à l'utilisation d'œstrogènes (c'est à dire les œstrogènes sans administration concomitante de traitement progestatif) a été associée à une importante, liée à la dose de risque de cancer de l'endomètre. Le risque peut être compensé de manière significative par l'ajout d'un progestatif, mais ne peut pas être complètement abolie. Avant d'entreprendre la thérapie d'oestrogène, les tests diagnostiques appropriés doit être réalisée chez les patients présentant des saignements vaginaux anormaux à exclure de l'endomètre tumeur maligne. La même chose s'applique si récurrente ou persistante saignements se développe au cours de la thérapie d'oestrogène.
